I am P.Seshu Vardhan, currently working as a full time research scholar at S.V National Institute of Technology, Surat., and pursuing Ph.D on the topic, natural inhibitors efficacy against SARS CoV-2. Due to pandemic, I wanted to be a part of global research to fight against CoVID-19 through phytomedicine and repurposing of drugs. I had received Pratibha award in the year 2019, by the Govt. of Andhrapradesh. And provisionally selected for DST (Department of Science and Technology) Inspire Fellowship. Professional career After my professional degree I was joined in the ROBO CLUB to learn Lego and Biloid robotics. After few months I worked as Robotics Trainer for high school students as
NXT Bots programmer. I got position in Target Telenetworks India Pvt Ltd a Vendor company of Reliance Jio. I worked as Optical Fibre and Network Engineer, to set right financial condition of my family. Later I was decided to continue my studies in Biotechnology. I have worked in the top most labs of India in Molecular Oncology Lab IIT Madras in the Department of Biotechnology under the Prof. Suresh Kumar Rayala. Here I have learned, how to do Cancer cells cultures and related experiments and Technics like Immuno- Precipitation Assay, Confocal Imaging, Transfection, q-PCR, Cell Viability Assay, and Cell Cycle Analysis etc. I Handled some of In-vivo Experiments on Zebra Fish along with my senior's PDF and Ph.D. students. I completed my project on “Studies on Targeted inhibition PAK1 with synthetic compounds (AZ13705339, FRAX597, IPA3) on Pancreatic Ductal Adenocarcinoma”. Some of the cell lines like Panc28, MiaPaCa2, Panc1, and some other gene knockdown cell lines. By exploring things in this lab makes me so excited to do more research on Cancer. When I read research papers on cancer I feel like I am doing average research as compared. Here we have limited sources and I want to explore more and learn advanced techniques. Since my class eight, I like Microscopes very much. This makes to stay in the lab for a long time to observe cells. With some experience on molecular oncology, I joined as Project Intern and continued as a project assistant for 11 months in Biochemistry lab at the Central University of Punjab, Bathinda. Under Dr.Shashank Kumar. My guide handling few governments funded projects on breast and colon cancer. I was handling breast cancer cell lines like MDA-MB-231, MCF7, T47D and colon cancer cell lines like HCT116 and HT-29. I did MTT assay more than 150 different herbal extracts and few synthetic compounds. We are going to publish 5 different Research paper at a time by the end of March-2019. Most of them are related projects with different compounds which screened out through molecular docking. Because of no Research papers, I missed lots of opportunities to study Ph.D. in foreign universities. At present, I am working on “Identification of Notch and stemness Inhibitors for MDA-MB-231 Breast Cancer stem cells. I have checked the protein expression fold of GAPDH, Hes1, Hey1, Caspase3, PAK1, SURVIVIN, Bcl2, CXCR4, MTH1, VEGFR2, Carbonic Anhydrase II, CHK1 through q-PCR. I have done molecular docking for all the protein receptors with some of the ligands founded in PubChem and ZINC database. Along with wet lab works, I have learned GROMACS, and Schrodinger’s to perform Molecular Dynamics Simulations.
Another project on Colon cancer cell lines was “Synergistic study on Natural drug formulations for Inhibiting multiple protein receptors of HT29 and HCT116”. We did another project similar to this project on MDA-MB-231 breast cancer. For this, we performed qPCR for VEGFA, RPL38, MYC, Nanog, MGMT, APE1, NFE2L2, SOD1, Oct4, Parp1, CD44, Gli1, EPCAM, BMI, NFE2L2. This project I experienced and performed Confocal and flow cytometry for colon and breast cancer cells probed with Rhodamine 123, Hoechst 3342, Annexin V, JC-1, H2DCFDA, different types of fluorescence dyes. Since my master’s degree to till now, I was handling these projects. So I submitted my thesis on “Targeted Inhibition of Protein Network and Cytotoxic Effect on Adenocarcinoma and Cancer Stem Cells”. During my B.Tech I submitted my Thesis Entitled “Molecular Identification of Dental Caries causing Microbiota through 16 rRNA sequencing & testing the Efficacy of Herbal Plant extracts of Indian traditional Medicine Bacterial isolates” under the guidance of Prof. Kaiser Jamil, Dean, School of Life Sciences, and Director – Centre for Biotechnology and Bioinformatics at Jawaharlal Nehru Institute of Advanced Studies, Hyderabad). And presented this paper at the International conference (ICBT- 2017) proceedings’ at JNTUH. I had published almost 4 research articles and more than 5 book chapters in different areas.
